A single dose of benzathine penicillin G is associated with similar serologic outcomes as the standard 3-dose regimen for patients with syphilis, including in those with late-onset disease or disease of unknown duration, according to study findings published in Clinical Infectious Diseases.

Syphilis is traditionally treated in the United States with 3 weekly intramuscular injections of benzathine penicillin G. However, limited contemporary evidence supports the necessity of 3 doses, and completion of the full regimen can be challenging. Therefore, researchers evaluated the relative effectiveness of benzathine penicillin G dosing strategies, as well as doxycycline, using surveillance data from multiple US jurisdictions.
